Backup vs. Replication

I am curious... is there a difference in the methodology for kicking off a backup vs. replication?

I have a system (Notes, Ugh) where the backup (no application awareness) works 100% of the time... lovely green checkmark.

Replication does not. While the policy has Application Awareness turned on, I have disabled it in the Application Aware Processing Options. The error I get is:

Failed to open attachment 'C:\ClusterStorage\Volume2\REPLICA VIRTUAL MACHINES\HYPER-V REPLICA\VIRTUAL HARD DISKS\2C705716-0401-467A-8B97-5C77B6519E90\NODENAME_C.VHDX'. Error: 'The process cannot access the file because it is being used by another process.'.'). Error code: '32774'.
Failed to get information about vhd disk 'C:\ClusterStorage\Volume2\REPLICA VIRTUAL MACHINES\HYPER-V REPLICA\VIRTUAL HARD DISKS\2C705716-0401-467A-8B97-5C77B6519E90\NODENAME_C.VHDX'

No, I've not opened a support ticket, and this evening my customer is going to Live Migrate the system to the storage location it is supposed to be in. We'll see if this helps.
