Howdy out there!
a customer wants to back up almost all O365 data - besides the personal onedrive(s). And preferably as default setting - not selecting the user and then deselect the OneDrive.
Is this possible at all?

It is recommended to create a backup repo per each data type:
- Exchange
- Sharepoint + Teams
- OneDrive
After you have the backup repos, create backup Jobs for each data type and target it to the dedicated Backup Repo.
If you want to exclude OneDrive, don't create a backup repo and backup job for the OneDrive Data.
I have written a post about choosing a specific data type for each job:

If you have already created a backup job, you can edit it to backup everything else except OneDrive.
Add the entire organization as the source and remove the option "OneDrive" it in this step:
- Exchange
- Sharepoint + Teams
- OneDrive

I'll create three separate repos as
a) path on one storage volume
or better
b) one volume per repo?
Selecting via datatype "User": I only see the possibility to select very single user - or am I missing something? Maybe start from scratch?
The customer already has a repo and setup "complete organization".

there are already separate jobs but they are pointing to the same path on the repo-volume.
Should or must there be different paths?

In my environment, we have one single large disk for multiple organizations.
- D:\REPO\ORG1\EXO
- D:\REPO\ORG1\SPO
- D:\REPO\ORG1\ODB
If I could rebuild it, I would create a vmdk per each organisations for data separation.
But it is not necessary for small deployments.
I don't have any experience with large deployments how it's done there.
And remove OneDrive from this selection:
