As mentioned in some of my other posts, I have 2 sites (HQ & Site-1) and approx 10 "mission critical" VMs that I want to have them on Site1 as both DR and 2nd backup location.
So what is the main difference between having a backup copy and a replica ?
Can I just power on a replica ? Or how do I "use" the replica as a VM ?

In simple way I would describe it like this:
The backup copy job allows you to have an independent backup chain at the secondary location.
The replication allows you to have another copy of a VM which can be started automatically in case of disaster.
Yes, you can manually fire up replica or set a planned failover, thus it will be started according to the schedule.

And you can use backups created by backup copy jobs as a source for replication jobs.
