We will be integrating DD620 for on-site backup + backup to tape (with offsite vaulting) for DR.
This is completely hypothetical, but If we send a weekly full backup to the DD620 (with Veeam compression/dedupe turned off) which is 5TB and then DD dedupes it down to 500GB. Will only 500GB be written to our tape media, or will the backup be "rehydrated" to 5TB and then written to tape?

First of all, please be aware that in the upcoming version of VB&R it will be required that the tape library should be attached specifically to the Veeam B&R management server, not the proxy or repository server.
Additionally, during “dedupe appliance to tape” copying process, all data will be definitely inflated to its original size.
