No. Assuming you're upgrading from 12.0 or 11, you should just upgrade in-place as you normally would. And if you're on 12.1 already, having successfully installed one using a previous ISO version, then you don't need the 20231206 ISO at all as it contains the same GA build (only the database upgrade script was updated, but you are passed that step already).

P.S. We've also added the ISO version history in our usual support KB article for [future] 12.1 cumulative patches, as we were getting questions periodically last year from customers worrying why an ISO was quietly updated and what exactly has changed.
Outside of ninja edits following a new version launch like the one above, we're repackaging ISO every time one of its components is updated (agents, plug-ins, appliances etc.) so that the product download always contains the latest and greatest.
