I want to upgrade our Veeam Backup & Replication environment to the newest version 6.1.
I've read the upgrade manual how to run an upgrade, but is it supported to upgrade from 5 to 6.1 or do I need to upgrade v6 first?

As per release notes it is supported if you have at least v5.0.1 or v5.0.2 running. If you are running with an older release you should contact support first.
And you should read the release notes carefully before upgrading which are included in the install package.
