All of my Veeam backup Servers are on the v12.1 latest edition, however, the database is still using the SQL Server Express 2012 edition.
What are the best practices and the supported migration path from this old database?

FYI: The SQL Server Express 2012 edition is no longer supported since 2022:

So it is better to migrate the database where practical.
