I have a physical B&R server and 3 VMs acting as proxies.
The proxy VMs are hosted by 3 different ESXi hosts with an iSCSI connection to the SAN hosting the VMware datastores where the VMs to be backed up reside.
Should I create an iSCSI initiator on each proxy to access the LUNs hosting the VMware datastores to allow direct access?
Is there any risk to compromise the integrity of the VMware data?

Yes, you should establish an iSCSI connection between the proxies and the SAN storage.
You will be on a safe side, since read-only mode is enough for proxy to retrieve the VM data during the backup process.
Anyway, using VM proxies with the direct SAN mode isn't recommended, as you will not get major benefits.
