New Windows File Server - best practice

Hello,

We have 2 physical servers with about 5 To of files. They replicate using DFS-R.
We are not satisfied with the current result and we are thinking about virtualizing this file server.

The result would be :
- one VM with Windows 2012 R2
- storage on Datacore SanSymphony V so it will replicate easily through the network

1 - Which advice can you give us regarding the storage?

We currently have 2 sites, 2 Datacore Servers (one on each site), 3 ESXi 5.5, and 1 Veeam 8 Proxy on each ESXi
2 - What is the best practice to backup with Veeam this VM and its data storage?
