I'm currently evaluating the product and have a few 400 GB VMs I need to test replication on. I have copied the initial replication to the USB drive and taken it to my COLO space, but I don't know how to get the files onto my datastore.
Has anyone else done this and care to share some pointers with me? I would certainly appreciate it!

You may use both FastSCP and Datastore browser to upload your VMs to the target datastore.

I have it connected directly to my ESX server but I can't seem to make it visible to the ESX server. I'm not a linux jedi unfortunately. It seems that would be the quickest way to get it over to the datastore, right?

The USB drive is not visible, because ESX server cannot read NTFS volumes. You should use vSphere Client's Datastore Browser or FastSCP functionality (built-in to Veeam Backup and Replication) to copy files to the datastore.
