For unstructured data backups, can the cache repository only be configured on a repository defined within the Veeam server? Or can it also reside on a share (network share) located on the Veeam server or on a proxy?
What are the most important constraints, and how is its required space calculated?

Your cache repository can be nearly any repository added to the backup server, with the exception of certain types like deduplication appliances and object storage. SMB shares are supported once they have been added as an SMB repository.
Our general recommendation is to choose a repository that is as close as possible to your file backup proxy. The file backup proxy is the component responsible for reading and writing data to the cache repository. For SMB shares, both your gateway server and the SMB share should be located in the same area as your file backup proxy.
For sizing, you can consult our best practice guide. Generally, the size of the cache should not exceed 4GB for most backup jobs. Please note, for file backup jobs with object storage as the target backup repository, we recommend allocating at least 5% of the size of your source data for the cache. Additionally, we suggest using SSDs for the cache in this scenario.
